Fluoride has been widely used to prevent dental caries, its toxicity(to the target cells) has been also published. This thesis was performed to understand the effect of sodium fluoride (NaF) on the viability and the morphological change of three different cell lines. Tested cell lines were L 929, NCTC 1469 and SiHa cell lines.
The cells were incubated with different concentration (0.0625%, 0.125%, 0.25%, and 0.5%) of NaF for 3 to 24 hrs (3,6,9,12, and 24 hrs). Cell viability was evaluated by the neutral red assay. Light microsopy was used to observe the morphological change after the incubation of 0.25% NaF for 3 and 6 hrs.
The results were as follows ;
1. The viability of L 929 cells was significantly decreased with the increase of NaF concentration and incubation time (P<0.05). The viability of NCTC 1469 cells appeared to be lower than 30% after the 6 hr incubation period. The SiHa cells, observed at least 50% in all tested concentration, suggested that these cells were the least influenced by the NaF.
2. In L 929 cells, the total cell numbers were decreased with NaF treatment. The cell membranes and cytoplasm were significantly destroyed with increasing of incubation time. In NCTC 1469 cells, the nuclear condensations and shrinkages were observed and the destruction of cell membrane and cytoplasm was also observed. Less cell numbers were counted than those of the L 929 cells. The SiHa cells appeared to be more resistant to morphological changes including cell numbers by NaF than other two cell lines. The results confirmed the previous viability test.
These findings suggested that there were some differences in fluoride sensitivity among three different cell lines, and the cell membrane and cytoplasm were damaged by fluoride.
