The objective of this research was to characterize the in vitro cellular behaviors of osteoblast-like-cell on titanium surfaces which were sterilized by either steam autoclaving, exposure to ethylene oxide gas, or exposure to ultraviolet light. Osteoblast-like cells in serum supplemented media were incubated on the standardized titanium specimens for up to 3 hours.
To evaluate cellular responses the number of cells attached was counted on the scanning electron micrograph which was taken at five randomly sampled sections from the surface of a specimen and the attachment area occupied by a cell was calculated using Image Analyzer.
The results were as follows:
1. The number of cells attached on ultraviolet light-treated surfaces was significantly more than that on both autoclaved surfaces and ethylene oxide-treated surfaces at any incubation period. But, there was no significant difference in t
he number of cells attached between autoclaved surfaces and ethylene oxide-treated surfaces.
2. There was no significant difference in the degree of cell spreading among autoclaved surfaces, ethylene oxide-treated surfaces and ultraviolet light-treated surfaces.
3. On the scanning electron micrograph there was little difference in early cellular morphology among 3 groups. But, as time went on, the degree of cytoplasmic extension on the ultraviolet light-treated surfaces was more advanced than that on the others.
These results indicated that ultraviolet light-treated surface had the highest amount of cell attachment and was not less than the others in the degree of cell spreading.
Therefore it was not suggested that the ultraviolet light treatment could lead to improvement in physico-chemical properties of the implant surface that might increase the clinical success rate of dental implants as well as reduce the time demanded to osseointegration.
